Three-dimensional construction of cultured cardiac myocytes and augmentation of their pulsatile ability by cell sheet manipulation utilizing temperature-responsive culture dishes

The utilization of temperature-responsive culture dishes for the construction of three-dimensional cultured cardiac myocytes was investigated. Primary chick cardiac myocytes were cultured on the substrates grafted with a temperature responsive polymer, poly(N-isopropylacrylamide) (PIPAAm), at 37°C, when substrates were slightly hydrophobic. Cross-sectional observation of the constructs revealed that three-dimensional cardiac tissues were fabricated by the unisotropic contraction and the fusion of cardiac myocyte sheets. The pulsatile ability of the cultured myocytes was augmented by the manipulation utilizing temperature-responsive culture dishes.
