Screening of circulating tumor cells in tumor-bearing mouse blood by a deterministic lateral displacement micro fluidic device

Abstract

Enrichment of CTCs in blood of tumor-bearing mice using a microfiuidic device by the deterministic lateral displacement method was demonstrated. Micro-posts structure for size based sorting was fabricated on Si substrate, and the screening for CTC detection was performed. Tumor-bearing mouse were prepared by inoculating tumor cells with GFP (green fluorescent protein) expression, and the CTCs were distinguished by its fluorescence. Before the sorting, no CTCs were detected in 1.5 million cells, while 12 CTCs were detected in 2.4 × 10 4 cells of larger cell section after the screening.
