Creation of hepatocyte spheroid array for high throughput screening

Abstract

The hepatocyte spheroid array was prepared by endothelial-hepatocyte co-culture on the poly(ethylene glycol) (PEG) brush surface micropatterned with plasma etch. Cell adherent domain sizes and their distances were important for effective spheroid formation. Live-dead staining and fluorescence immunohistochemistry revealed that hepatocyte spheroids underlaid with endothelial cells kept high viability and well-maintained hepatocyte specific function for long period.
